چکیده
Abstract Purpose Coral degradation is a worldwide ecological problem. Bacterial diseases are great danger to coral health. The pathogenic bacterium Vibrio alginolyticus XSBZ14 isolated from diseased had been identified as the of Porites andrewsi White syndrome (PAWS) in Xisha Archipelago on transmission experiment. To date, molecular mechanism by which this pathogen causes disease unknown, and diagnostics for caused have not developed. In an effort restore damaged ecosystems South China Sea, efforts underway transplant flat-branch shore corals. There therefore urgent need further develop specific rapid detection methods V. order prevent epidemic ensure successful implementation compilation transplants. Methods At first, low sequence identity single-copy S2 was selected genome in-house Perl script. Using designed primers, four different types standard curves were subsequently plotted accurate quantification strain samples (DNA, bacterial suspension, tissue, seawater). Then, use infect Galaxea fascicularis test culture water during week. Results method bacteria RT-PCR established. limit (LOD) 0.88 pg/reaction (0.44 pg/μL) DNA, 2 CFU/reaction (1000 CFU/mL) 20 seawater XSBZ14, respectively. addition, according results RT-PCR, could survive week, also be detected its reared seawater. Conclusion These indicated that robust tool pathogen, very useful PAWS epidemiological survey pathogen-free transplantation Sea. And other species their habitats might act important reservoir mediated horizontal reefs.

متن کاملA novel method for real time quantitative RT-PCR.
A novel approach to quantitative reverse transcriptase polymerase chain reaction (QC RT-PCR) using real time detection and the 5' nuclease assay has been developed. Cystic fibrosis transmembrane transductance regulator (CFTR) target mRNA is reverse transcribed, amplified, detected, and quantitated in real time. A fluorogenic probe was designed to detect the CFTR amplicon. Relative increase in 6...
متن کاملEndophthalmitis caused by Vibrio alginolyticus.
Vibrio alginolyticus is a facultative anaerobic gram-negative bacillus found in normal marine flora. Ocular infections induced by V. alginolyticus are extremely rare. We report a case of endophthalmitis caused by V. alginolyticus to draw attention to V. alginolyticus infections following ocular injuries.
متن کاملBiodesulfurization of Dibenzothiophene by a Newly Isolated Thermophilic Bacteria Strain
Microbiological analyses of soil chronically exposed to petroleum complex compounds of some oil springs in south of Iran resulted in isolation and purification of a new native thermophilic strain which is capable to desulfurize petroleum sulfur compounds by 4-S mechanism. Dibenzothiophene (DBT) was selected as a complex sulfur compound model and many experiments were done to identify the me...
متن کاملDetection and quantitation of bovine respiratory syncytial virus using real-time quantitative RT-PCR and quantitative competitive RT-PCR assays.
A single tube, fluorogenic probe-based, real-time quantitative reverse transcription-polymerase chain reaction (Q-RT-PCR) assay was developed for detection and quantitation of bovine respiratory syncytial virus (BRSV) using BioRad's iCycler iQ. Real-time Q-RT-PCR was compared with quantitative competitive RT-PCR (QC-RT-PCR) and viral titers. Viral mRNA levels were measured in BRSV-infected bovi...
